New Thiazole Derivatives For Treatment Of Alzheimer’s Disease

Abstract
Alzheimer's disease (AD) is a neurodegenerative disorder mostly influencing the elderly and causes death due to dementia. The main pathogenic feature connected with the progression of this multifactorial disease is the weakening of the cholinergic system in the brain. Cholinesterase (ChEs) inhibitors are recognized as one of the most promising targets in the treatment of AD. The inhibition of both enzymes, AChE and BChE, has been approved as an appropriate therapeutic strategy to reduce the symptoms of disease and prevent disease progression; as well. Thus, in this study seven new thiazole-piperidine/piperazine derivatives were designed and synthesized to able to have good inhibition profile as cholinesterase inhibitors. The structures of synthesized compounds were confirmed via 1H and 13C NMR spectroscopic methods. The anticholinesterase activity of final compounds was performed by using modified Ellman’s colorimetric method. According to enzyme inhibition assay, none of the compounds displayed significant inhibitory activity against BChE enzyme. On the other hand, compounds 3f and 3g showed promising inhibition potency against AChE enzyme with IC50 values of 0.1425±0.006 µM and 0.1384±0.005 µM, respectively.
